Caffeine attains the same concentration in gonadal tissue as in the circulating plasma and may thus reach levels of up to 0.05 mM for very heavy consumers (43). In vitro mutagenicity studies in which positive results are obtained generally involve concentrations at least 100 times greater and are therefore at or above the lethal concentration for humans. At these high concentrations, DNA repair is inhibited, an effect attributable to its purine analog structure. Caffeine may thus be considered a weak mutagen with no significance for humans.
