Antigen structure

The CD52 antigen is well defined at a genetic and structural level, but its function is currently unknown. Encoded by a single gene on chromosome 1, it is a remarkably small glycoprotein, having only 12 amino acids (Figure 1). Attached to Asn3 is a complex carbohydrate consisting of sialylated polylactosamine units with a tetra-antennary fucosy-lated mannose core. At the C-terminus is a glycosyl-phosphatidylinositol (GPI) anchor. The CAMPATH-1 epitope includes the C-terminal amino acids and part of the GPI anchor. This implies that the antibodies bind close to the cell membrane, which explains why they are so good for cell lysis. Other antigens with a similar structure to CD52 include CD24 and its mouse homologue J5 (heat-stable antigen).
