Two signals are required for optimal T cell activation. Specificity is provided by the interaction of the T cell receptor (TCR) with major histocompatibility complex (MHC)-peptide but a second, antigen-unrestricted costimulatory signal is also required. The B7 family of proteins, B7-1 (CD80) and B7-2 (CD86 or B70) provide the major costimulatory signal for T cell activation and clonal expansion via their interaction with CD28. Engagement of the TCR in the absence of B7 costimulation results in T cell clonal anergy.
