The conserved li chain and polymorphic MHC class II molecules are selectively coexpressed in a tissue-specific fashion in conventional antigen-presenting cells such as B cells, macrophages and dendritic cells known to be responsible for initiating CD4+ T cell responses. Ii chain and MHC class II gene expression are coordinately upregulated in response to cytokines such as interferon (IFN) and interleukin 4 (IL-4). Many of the transcriptional regulatory elements mapping to their promoter regions appear largely conserved, but control of gene expression differs in at least one important aspect because Ii chain transcripts are far more abundant than MHC class II, probably reflecting in part the actions of a transcriptional enhancer located in the Ii chain first intron.
