ICC methods can be divided into four groups:

The direct method (see Figure 1) Here a labeled antibody (known as the primary antibody) attaches directly to the target antigen in the tissues. An inherent problem with this system is that unwanted antibodies directed against tissue antigens are also labeled and can attach to the tissue section thus producing high backgrounds. The use of mAbs can reduce this contamination.
