The actions of IL-8 on neutrophils are diverse (Figure 4): chemotaxis, transendothelial migration with concomitant shedding of leukocyte adhesion molecule 1, induction of lysosomal enzyme release, respiratory burst, shape change and generation of superoxide anions. Moreover, IL-8 induces chemotaxis of T and B lymphocytes, IL-2-activated natural killer cells, basophils and eosinophils if primed with cither IL-3 or granulocyte-macrophage colony-stimulating factor (GM-CSF). IL-8 inhibits IL-4-induced immunoglobulin E (IgE) production by normal B lymphocytes and inhibits in vitro colony formation from human bone marrow progenitor cells.
IL-8 has profound effects on nonleukocytic cells. IL-8 is haptotactic (induces cell migration to cell surface- or matrix-bound gradient), and promotes adhesion and proliferation of melanoma cclls. It inhibits collagen expression in synovial fibroblasts and enhances cytomegalovirus replication in lung fibroblasts. Furthermore, IL-8 induces the migration of human umbilical vein endothelial cclls in vitro and angiogenesis in rat cornea.
