Granzymes are synthesized as pre-propeptides. Following cleavage of classical leader peptides, they are transported through the Golgi, then targeted to the lysosome-like granules by binding to mannose-6-phosphate receptors. Immediately prior to packaging, cleavage of a remaining N-terminal dipeptide by dipeptidylpeptidase I results in refolding of the mature N-terminus into a cleft in the molecule, approximation of the 'charge-relay' residues and acquisition of protease activity. All granzymes are basic molecules (pi in the range 8-11) and are retained in cytolytic granules (pH ~5.5) electrostatically bound to chondroitin sulfate proteoglycans which are present in high concentration.
