The cDNA encoding interleukin 11 (IL-11) was originally isolated from a primate bone marrow-derived stromal fibroblast cell line based on its mitogenic activity on an IL-6-dependent mouse plasmacytoma cell line. The same cDNA was also isolated independently by its ability to inhibit adipogenesis. Subsequent in vitro, in vivo and clinical studies have shown that IL-11 is a multifunctional cytokine important in hematopoiesis. Mouse and human IL-11 receptor (IL-11R) cDNAs have been isolated by hybridization using degenerate oligonucleotides for the WSXWS motifs of hematopoietiin receptors and an enhancer-trap method. Sharing of the common gpl30 signal transducer among IL-11, IL-6, leukemia inhibitory factor (LIF), oncostatin M (OSM), ciliary neurotropic factor (CNTF) and possibly cardi-otropin 1 (CT-1), at least in part, the biological redundancy and pleiotropy of these cytokines. In vitro and in vivo studies have implicated IL-11 as a multifunctional growth factor with therapeutic potential in enhancing thrombopoiesis, protecting gastointestinal injury and modulating immune response. Like many other hematopoietic growth factors its clinical efficacy should be interpreted with caution and may be further improved by combination with other hematopoietic growth factors.
