Lymphoid and epithelial 78 cells

The anatomical representation of y8 cells varies strikingly. In adult mice, y8 cells account for - 1-3% of T cells in spleen and lymph nodes, 10-50% of intestinal intraepithelial T cells, the majority of reproductive epithelial T cells, and &95% of resident epidermal T cells. In humans, y8 cells usually account for from 0.5% to 15% of T cells in peripheral blood, are variably represented in the gut epithelium, but are not obviously present in the skin. In chickens, y8 cells frequently comprise 30% of peripheral T cells, and can constitute the majority of gut intraepithelial T cells. In ruminants, 30-80% of blood T cells are 78 cells.

A high proportion of 78 cells are double negative (lack surface CD4 and CD8) (see Tables 3 and 4), reflecting the fact that 78 cells generally do not pass through positive selection by class I or class II MHC. Many gut 78 cells are CD8+, but express the CD8ota homodimer that may be unrelated to recognition of class I MHC.

Murine epidermal 78 cells can recognize kera-tinocytes, and gut 78 cells may recognize gut epithelial cells. Epithelial cells acting as antigen-presenting cells would be relatively immobile, providing for few encounters with T cells. Thus, if the T cells are to be activated, those few encounters must have high chance of presenting a 78 TCR ligand. This concept provoked the idea that intraepithelial 78 T cells would have limited antigen specificity, strikingly borne out in the murine skin and reproductive epithelia.
