The PKC pathway

DAG, the other product of PLC activation, along with cofactors such as Ca2~ and phospholipids, activates protein kinase C (PKC), a family of serine/threonine kinases that phosphorylate a number of substrates, initiating the PKC pathway. PKC has been implicated in the control of many signaling pathways, including p2T"s and the MAP kinases such as ERK, and c-JUN N-terminal kinases (JNK 1/2). The role of specific PKC isozymes is poorly understood, although PKCe and PKC0 can regulate the transcription factors AP-1 and NFAT in leukemic T cells, and a recent study indicates that PKCfl may have a special role during antigen activation of T cells. Phorbol esters activate many forms of PKC, however it is now clear that many effects of phorbol esters on T cells that were previously attributed to PKC are in fact Ras mediated. In vitro studies have shown that the PKC isozymes 8, e, tj, A and £ arc activated by D-3 phosphoinositide lipids. PKCf has also been suggested as a direct target for ceramide and thus may play a critical role in sphingomyelinase signaling.
