Graft rejection is another area in which the immunomodulatory effects of PAF appear to be important. PAF is known to be released from renal allografts during hyperacute rejection and the PAF antagonist BN 52021 increases cardiac, liver, and skin allograft survival in rats, acting synergistically with azathio-prine and cyclosporine. The PAF antagonist also prevents cyclosporine induced nephrotoxicity without altering the immunosuppressive effect of the drug. In addition, treatment with BN 52063 in combination with cyclosporine increases the immunosuppressive effect of the latter.
