Accuracy Reproducibility And Precision Of Galios Genotyping

The performance of GALIOS™ is demonstrated by genotyping the SNP at position 655 (A655G) within the human MLH1 (mutL homolog 1) gene.[11] The hMLHl protein functions as a mismatch repair protein and helps to maintain the fidelity of DNA during replication. The A655G transition within the hMLHl gene replaces isoleucine at position 219 by valine (I219V) in the hMLH1 protein and is associated with the inherited familial cancer syndrome of hereditary nonpolyposis colorectal cancer (HNPCC).[12]

To compare the accuracy of the GALIOSTM genotyping technology to a conventional genotyping method, 60 genomic DNAs (gDNAs) were analyzed in parallel by

1) PCR amplification and subsequent restriction fragment length polymorphism analysis (PCR-RFLP) and by

2) GALIOSTM.

For PCR-RFLP, a 191-bp PCR product encompassing the A655G polymorphism of the hMLHl gene was amplified from each gDNA. The antisense primer was designed such that a Clal restriction site (AT/CGAT) was introduced into the 191-bp PCR product upon amplification from allele A but not from allele G. Consequently, restriction digests of the amplimers with ClaI analyzed by agarose gel electrophoresis revealed distinct restriction fragment patterns for the three hMLH1 genotypes (Fig. 2A and B).
