Resuspend cDNA targets in 5 x SSC/0.1% sodium dodecyl sulfate (SDS) and denature at 99°C for 2 min. Add 50 mL of solution to a prewetted glass slide (2 x SSC, 0.1% SDS), cover with a glass cover slip, and place in a moist chamber and incubate at 65°C for 12-18 hr. Then wash in 2 x SSC/0.1% SDS for 2 min, 1 x SSC, 0.2 x SSC, and 0.05 x SSC sequentially for 1 min each before scanning.

Perform hybridizations in 6 x SSC, 0.3% SDS, and 5 mL of the amplified DNA. Heat-denature then wash slides for 15 min at room temperature in 3.3 M tetramethyl ammonium chloride (TMAC).
