Introduction

Gaucher disease, the most common lysosomal storage disorder, is caused by deficient p-glucocerebrosidase activity and results in progressive accumulation of the substrate in macrophages, leading to hepatosplenomegaly, anemia, thrombocytopenia, skeletal lesions, lung disease, and sometimes neurological involvement. Enzyme replacement therapy (ERT) has proven to be safe and effective. Other strategies including bone marrow transplantation, substrate reduction, enzyme chaperones, and gene therapy have been variously attempted.
