Reversed line blot (RLB) hybridization is a specialized form of allele-specific oligonucleotide assay that provides a robust method for the molecular characterization of genes with numerous nonsynonymous variants. In most highly refined form, these assays rely upon highly multiplexed PCR reactions containing biotinylated primers to provide a substrate for nonradioactive detection systems. Reversed line blot technology involves mechanized covalent attachment of activated primary amine-conjugated oligonucleotides to carboxylated nylon membranes or BSA. Printed membranes are stored or sold dry in preparation for hybridization. Lines are visualized colorimetrically after hybridization using streptavidin horseradish peroxidase incubation then developed using a tetramethylbenzidine derivative and hydrogen peroxide. For rapid prenatal diagnosis of cystic fibrosis (CF), p-thalassemia, and other hemoglobinopathies, this technology is especially applicable. Commercial instruments are available to provide greater uniformity for the hybridization and color development.
