Capillary gel electrophoresis requires smaller amounts of sample compared to slab gels (nanoliters compared to microliters) to avoid excessive band broadening.[4] Loading is automated, and injection volume can be controlled and standardized by varying the force and time. Samples can be loaded in a number of ways including hydrody-namic or electrokinetic injections. For hydrodynamic loading, the capillary is subjected to a pressure difference between its extremities. The pressure difference can result from gravity, application of pressure inside the sample vial, or aspiration of the sample through the capillary tube. Hydrodynamic injection produces more reproducible sample loading. For electrokinetic injection, the capillary tip is dipped into the sample for a fixed time and a potential is applied. Electrokinetic injection results in the best flow profiles but can give a biased sample because more mobile ions can be preferentially introduced.
