Signal Shift

A signal shift during the day of the screen is one frequently faced problem. In screens with long plate-handling or incubation periods, plates prepared during 1 to 4 h are used for more than 24 h. Cell plates used later after seeding can have higher cell numbers or different medium compositions compared to those plates used earlier. In addition to growth arrest and hypothermia, the seeding of plates for one day with two to four different cell concentrations can help to reduce the shift effect. An automated plate seeding procedure reflecting the time frame of the assay procedure is ideal.
